Yes. Easy, but didn't worry about draining cooling line and cooler. Most important thing to remember is to open the fill plug before the drain plug (to ensure you can refill it, once it's drained).

Im going to be using Royal Purple Synchromax Manual Transmission Fluid. do you think it matters if i don't drain the cooling line and cooler? somewhere i saw that you have to have the car on while refilling it. is that the right way to do it?
thanks

Bad idea on the RP Synchromax... lots of negative reviews on it.

Mobil 1 ATF is an upgrade over the stock fill.

Other great choices are Redline ATF D4, or Amsoil ATF.

Check the viscosities of some of your choices. You want something with similar properties as the Dextron fill it came with. Some of the heavier choices can cause the synchros to drag until the fluid is all warmed up. Primary reason I'm running Mobil 1.

Per the Service Manual"
MERCON® V Automatic
Transmission Fluid
XT-5-QM (or XT-5-QMC) (US);
CXT-5-LM12 (Canada)

I use Amsoil.

1. With the vehicle in NEUTRAL, position it on a hoist. Refer to Section 100-02
2. Remove the drain plug and drain the transmission.
3. Clean the drain plug. Apply threadlock and sealer to the plug threads and install the drain plug.
 Tighten to 27 Nm (20 lb-ft).
4. NOTE: Before removing the fill plug, clean the area around the fill plug.
Remove the fill plug.
5. Using a oil suction gun, fill the transmission with the specified fluid until it is even with the oil fill plug
hole.
6. Install the fill plug. Start the engine and shift the transmission through all the gears to eliminate any
trapped air in the transmission fluid cooling system.
7. Remove the fill plug.
8. Using a oil suction gun, fill the transmission with the specified fluid until it is even with the oil fill plug
hole.
 Transmission capacity is 3.96L (4.18 qt).
9. Apply threadlock and sealer to the plug threads and install the fill plug.
 Tighten to 25 Nm (18 lb-ft).

I just did mine a couple weeks ago and used the amsoil signature series atf multi vehicle and there was a noticeable difference in smoothness of gear engagement.

If you have a TP I would recommend disconnecting the trans cooler lines from the transmission and letting them drain so you can, for the most part ensure you have all the same fluid in there.

Norton has it correct about draining the cooling lines. Such a small amount will not make a difference. The lines and cooler might hold 1/2 pint.

One more point I don't see posted here, if your not using a lift, make sure the car is level, reference refilling.

I was surprised at the noticeable difference Mobile 1 made. So this is worth the effort

Transmission fluid without cooler 3.6 quarts (3.4L)
Transmission fluid with cooler 4.1 quarts (3.9L)
Motorcraft® MERCON® V

The TREMEC 6060 6-speed manual transmission on your Shelby GT500 uses
MERCON® V automatic transmission fluid. Make sure that the correct MERCON® V ATF automatic transmission fluid is used as indicated on the label on your transmission.

Service refill capacity is determined by filling the transmission to the bottom of the filler hole with the vehicle on a level surface. For vehicles with a transmission cooler, the engine must be running, the clutch pedal must be released, and the transmission must be in neutral to perform the service refill.

I haven't tried this, but if I wanted to flush the old oil out of the cooler and lines, I would drain the old oil from the trans, refill with fresh oil and then disconnect the return line from the cooler at the trans. This part is going to take 2 people, start the engine and catch the oil from the return line and shutoff engine after you see about 4 ounces of new oil. Reconnect oil return line and add oil to trans until you reach the correct level.

Caution: Since I haven't tried this procedure, I would be on alert when the car first fires up until I witness the volume and pressure of the stream coming out of the return line. The pump gears don't look very large but unless you don't care if you get hosed down with ATF, I wouldn't go above idle and hopefully you chose your help do to their cat like reflexes and he or she will shut down the engine at the very second they hear your girly screams.

This may help.

I've tried RP, Amsoil and Mobil 1.

Mobil 1 is the best one in my experience.

I noticed a significant improvement by dumping the OEM and going Amsoil.
I'm sure the Mobil 1 and RP and great too.
